Olympiakos are locked on seven points with Benfica but are ahead of the Portuguese side courtesy of having the better head-to-head record.
That means three points on Tuesday will make it impossible for Benfica to advance.
The loss of Mitroglou is a major blow, but the Greek side head into this key encounter boosted by a weekend league win over Asteras Triploi.
Coach Michel resisted the temptation to rotate his squad in that fixture, but has no fears that his side will lack any freshness on Tuesday.
But he believes a much-improved performance will be required.
He told a press conference: "My selection on the weekend was not so much a matter of looking for freshness as looking for success and continuity.
"But I was disappointed with how many times we lost the ball when we had possession.
"It is true maybe we did have our minds on Tuesday, especially as it is a competition that has the eyes of the world on it and that can affect players.
"But on Tuesday we will need to be fresher, faster, more aggressive and end as winners."
Anderlecht, meanwhile, are rooted to the bottom of the group and have no hopes of even securing a Europa League spot having picked up just one point from their five games thus far.
It means Anderlecht will finish bottom of their group for the seventh time in the Champions League, and there is a good omen for their hosts in that the Belgian club have not won an away fixture in the Champions League proper since the 2005-06 campaign.
Olympiakos, by contrast, have won eight of their last 11 home games in the group stage.Mitroglou out as Greeks go in search of last-16 berth
The striker remains sidelined with medial ligament damage ahead of the game in Piraeus, with Michel's side knowing a win will send them through.
Olympiakos have their sights set on the knockout stages of the Champions League and will try to get there by beating Anderlecht on Tuesday.
The Greek side are on a seven-game unbeaten run at home in all competitions as they entertain the team bottom of Group C knowing that a place in the last 16 is within touching distance.


Anderlecht have struggled in this year's competition. Indeed, one point from five games means the Belgian champions cannot qualify for the Europa League and will finish bottom of a group for the fifth time.
Javier Saviola and Kostas Manolas were both rested ahead of the clash, while Gaetan Bong and Michael Olaitan were left on the bench.
Having been knocked out of the Coupe de Belgique to Westerlo, Anderlecht bounced back to winning ways with victory over Waasland-Beveren on Saturday.
Anderlecht sit third in the Pro League, but it is on the European stage where they have struggled. The Belgium side travel to Greece with no win away from home in their last six Champions League fixtures.
John van den Brom also has one player out through injury – Matias Suarez – while 20-year-old striker Massimo Bruno will miss the match through suspension.
